Revue d'histoire de la pharmacie - Année 1989 - Volume 77 - Numéro 281 - Pages 175-184Quinine, Antimalarials and resistance of Plasmodium. The celebration of the bicentennial of the birth of J. Pelletier provides the occasion to remember the history of the discovery and the use of quinine and other antimalarials which are chemically related. Following the isolation of the alkaloids of Cinchona by J. Pelletier, followed by chemical analysis resulting in the total synthesis of quinine, research into antimalarial substances was developed. The conditions of war during both world conflicts impeded the normal furnishing of supplies to occidental countries and gave a boost to research. Thanks to the original experimental models of malaria, chemists were able to develop 8-amino quinolines, acridines, 4-amino quinolines, methanol- phenanthrenes and methanol-quinolines whose antimalarial properties were more powerful and more long-acting than those of quinine. But the rapid appearance of strains of P. falciparum resistant to the antimalarials available in the bloodstream is a serious handicap and requires continuing research and the development of new original chemical series.
